چکیده

Combining reverse transcriptase-polymerase chain reaction (RTPCR) and molecular cloning technologies, we have investigated the expression of homeodomain proteins in chicken gizzard tissues undergoing differentiation. We demonstrate that homeodomain encoding transcripts are found in developing chicken gizzard. An RT-PCR generated 117 bp homeodomain DNA probe was used to screen a gizzard library and resulted in the identification of ~60 potential homeodomain encoding cDNAs. Sequence analysis of the two longest clones identified them as Hox b4 and Hox b5, paralogues of Drosophila Deformed (Dfd) and Sex combs reduced (Scr), respectively. Northern blotting analysis demonstrated an increase in the Hox b4 and Hox b5 mRNAs just prior to the onset of smooth muscle cell differentiation as judged by the expression of the smooth muscle γ-actin gene. Thus, Hox b4 and Hox b5 display the appropriate temporal expression pattern to be involved in the development/differentiation of the chicken gizzard.

Analysis of HOX gene expression patterns in human breast cancer.

HOX genes are highly conserved transcription factors that determine the identity of cells and tissues along the anterior-posterior body axis in developing embryos. Aberrations in HOX gene expression have been shown in various tumors. However, the correlation of HOX gene expression patterns with tumorigenesis and cancer progression has not been fully characterized. Here, to analyze putative cand...

Homeobox genes, fetal wound healing, and skin regional specificity.

Why do adult wounds heal with scars but fetal wounds heal without scars? Why do fetal tissues regenerate while adult tissues only repair damage? Why do some parts of adult skin produce scars more than other parts? Why in some genetic diseases or infectious diseases, are only particular skin regions a¡ected, forming speci¢c anatomical patterns? How are these temporal and spatial di¡erences in th...

Key functional role and lineage-specific expression of selected HOXB genes in purified hematopoietic progenitor differentiation.

Although it is well established that homeobox (HOX) genes play a key role in normal human embryogenesis, the expression and function of HOX genes in normal hematopoiesis is largely unknown. We have investigated by reverse transcriptase-polymerase chain reaction the mRNA expression of HOXB cluster genes (3' to 5' position in the cluster: from HOXB2 through B9) in 72% to 88% purified hematopoieti...

NF-Y cooperates with USF1/2 to induce the hematopoietic expression of HOXB4.

The transcription factor homeobox B4 (HOXB4) is preferentially expressed in immature hematopoietic cells and implicated in the transition from primitive hematopoiesis to definitive hematopoiesis as well as in immature hematopoietic cell proliferation and differentiation. We previously identified Hox response element 1 (HxRE-1) and HxRE-2/E-box as 2 critical DNA-binding sites of the HOXB4 promot...
